Here is the only succesful 2wd bike ever made so far. It has competed in Enduro races and has an advantage in the sand. It's made by Yamaha Belgarda (Italian subsidiary) and the front wheel is driven by a simple (relatively) hydraulic system with oil.

Anyone ever ride a ROKON? Man, whata beast! You can even fill the tires full of water if you think you need to. I rode one years ago without water in the tires and it was quite a treat. You don't steer 'em, you just aim it in the general direction you wanna go.
Do you see how much these things cost? Would you pay five grand for a bike with less than 10 horsepower? Man, I thought that one of these would be cool as a cheap substitute for a real dirtbike, but seeing that $4875 price-tag sure shot that idea to pieces.
Well, when you look at the hauling and towing capacities (1000 and 3000 lbs, respectively), the 60% grade-climbing ability, the 9 gallons of extra fuel or water storage in the wheels, the super light weight (only 208 lbs dry) , and its ability to friggin' float, you start to get the idea that this is something not really designed for crazy, bark-eating fun. It's a utility vehicle for outdoorsy type stuff.
